New Peer-Reviewed Clinical Study Shows Trpti™ Supports the Body’s Natural Incretin Response

Published in Frontiers in Endocrinology, the research provides further insight into how Trpti™ supports metabolic health, weight management, and healthy energy levels.

Saanroo today announced the publication of a new human clinical study evaluating Trpti™, its bioavailable oleoylethanolamide (OEA) ingredient powered by LipiSperse® technology. Published in the peer-reviewed journal Frontiers in Endocrinology (Impact Factor: 5.7), the study provides new insight into how Trpti supports nutrient sensing and the body’s natural incretin response, further strengthening the scientific foundation behind the ingredient for metabolic health and healthy weight management.

The fixed-sequence, single-blind, placebo-controlled, three-period crossover study enrolled 37 healthy adults with a body mass index (BMI) of 25.0-34.9 kg/m². Volunteers received a single dose of a placebo, 150 mg Trpti, or 300 mg Trpti following an overnight fast. Blood samples were collected over 8 hours, with standardized meals provided at 15 minutes and 4 hours post-dose. Time-course responses of Glucagon-Like Peptide-1(GLP-1), Glucose-dependent Insulinotropic Polypeptide (GIP), Dipeptidyl peptidase-4 (DPP-4), insulin, glucose, and glucagon were analyzed.

The study found that:

  • A single 300 mg dose of Trpti significantly increased post-meal GLP-1 and GIP responses in overweight adults compared with placebo. The effect was observed rapidly, within 15 minutes of ingestion, and recurred after subsequent meals.
  • Significant changes in circulating DPP-4 concentration were also observed at later time points, suggesting a potential association between Trpti supplementation and modulation of serum DPP-4. DPP-4 is an enzyme that degrades incretin hormones such as GLP-1 and GIP. By inhibiting DPP-4, Trpti may prevent the degradation of GLP-1 and GIP, allowing them to act with optimal efficacy.
  • Participants receiving Trpti consumed significantly less of a standardized meal than those receiving the placebo.
  • Trpti was well tolerated, with all safety biomarkers remaining within normal clinical ranges.
  • The authors concluded that a single 300 mg dose of Trpti enhances post-meal GLP-1 and GIP responses and demonstrates rapid engagement of GLP-1, GIP and DPP-4, supporting further investigation of its role in nutritional strategies targeting metabolic health.

This publication complements Saanroo’s previously published clinical research on Trpti.

In a 12-week randomized, placebo-controlled clinical trial published earlier this year, Trpti significantly increased the abundance of Akkermansia muciniphila and Faecalibacterium prausnitzii, two beneficial bacterial species associated with gut and metabolic health. The study also demonstrated statistically significant weight reduction in participants with a BMI below 35 compared with placebo, while supporting gut barrier integrity and healthy immune signaling.

Together, these published human studies demonstrate that Trpti supports healthy weight management through multiple complementary pathways, including appetite regulation, gut microbiome health, gut barrier function, and the body’s natural incretin response.
